ABSTRACT

【Objective】 To explore the mechanism of macrophage-inducible C-type lectin (Mincle) and microinflammatory state in uremia. 【Methods】 SD rats were randomly divided into uremia group and sham operation group. The morphology and permeability of intestinal tissue, the morphology of intestinal tissue and macrophages were observed by transmission electron microscope, the expression of Mincle was detected in intestinal tissue sections, and the expressions of Toll-like receptor 4 (TLR-4) and NF-kappa B (NF-κB) protein on the surface of macrophages were detected by Western blotting. After the plasma was separated, the levels of endotoxin, C-reactive protein (CRP), interleulin-6 (IL-6) and tumor necrosis factor-α (TNF-α) were detected by Limulus lysate dynamic turbidimetric assay, and enzyme-linked immunosorbent assay (ELISA). The data were analyzed with IBM SPSS19.0 software. 【Results】 The expression of Mincle in the jejunum, ileum, and colon in uremia group was higher than that in sham-operation group (P<0.05). The expressions of TLR4 and NF-κB protein significantly differed in the ileum, jejunum and colon in uremia group (P<0.001). The levels of endotoxin, CRP, IL-6, and TNF-α were significantly increased in uremia group compared with sham-operation group (P<0.05). 【Conclusion】 In uremia, Mincle on the surface of intestinal macrophages increases and further through TLR4/NF-κB pathway mediates the transformation of intestinal macrophages to M1 type, releasing inflammatory products and causing systemic microinflammation.

ABSTRACT

OBJECTIVE To explore the improvement effect and potential mechanism of total flavonoids from Alpinia zerumbet on gastric mucosa injury induced by absolute ethanol through microRNA-146a-5p (miR-146a-5p). METHODS Using human gastric mucosa GES-1 cells as objects, the acute gastric ulcer model was established by absolute ethanol; based on the investigation of the effects of different concentrations of total flavonoids from A. zerumbet on cell activity and the selection of action concentration, the relative expression level of miR-146a-5p in GES-1 cells was detected, the protein expressions of tumor necrosis factor (TNF) receptor-associated factor 6(TRAF6), nuclear factor-κB p65 (NF-κB p65) and TNF-α were detected, and the levels of interleukin- 1β (IL-1β), IL-6 and prostaglandin E2 (PGE2) in cell supernatant were determined. The targeting relationship between miR-146a- 5p and TRAF6 was verified; the effects of overexpressed miR-146a-5p and TRAF6 knockdown on the levels of IL-1β, IL-6 and PEG2 in supernatant of model cells as well as the effects of miR-146a-5p knockdown on anti-gastric ulcer effect of total flavonoids from A. zerumbet were observed. RESULTS Compared with the blank group, the relative expression of miR-146a-5p in cells and the level of PGE2 in cell supernatant were decreased significantly in the model group (P<0.01), while the protein expressions of TRAF6, NF-κB p65 and TNF-α in cells and the levels of IL-1β and IL-6 in cell supernatant were increased significantly (P< 0.01). Compared with the model group, the relative expression of miR-146a-5p in cells and the level of PGE2 in cell supernatant were increased significantly in model+A. zerumbet total flavonoids (60 mg/L) group (P<0.01), while the protein expressions of TRAF6, NF-κB p65 and TNF-α in cells and 82260767) the levels of IL-1β and IL-6 in cell supernatant were decreased significantly (P<0.05 or P<0.01). There was a targeted relationship and a negative correlation between miR-146a-5p E-mail:3113836821@qq.com and TRAF6. After overexpression of miR-146a-5p or TRAF6 knockdown, the levels of IL-1β and IL-6 were decreased significantly in cell supernatant, while the level of PGE2 was increased significantly (P<0.05). After miR-146a-5p knockdown, the levels of IL-1β and IL-6 in cell supernatant and the protein expression of TRAF6 in cells administered with total flavonoids of A. zerumbet were increased significantly, while the level of PGE2 was decreased significantly (P<0.05). CONCLUSIONS Total flavonoids of A. zerumbet can improve the gastric mucosa injury induced by absolute ethanol. The mechanism may be related to up-regulating the expression of miR-146a-5p, inhibiting the expression of TRAF6, and further inhibiting the secretion of related inflammatory factors.

ABSTRACT

Objective:To construct breast cancer organoid culture system and conduct histological characterization and preliminary radiobiological characteristics study.Methods:Different molecular types of breast cancer cell lines and patient-derived tumor cells were cultured in vitro to form breast cancer organoids and characterize their tissue structure. In addition, Ki-67, ER, PR and Her2 markers were evaluated by immunohistochemical staining. Breast cancer organoids were irradiated with 4 Gy and 8 Gy. The numbe and diameter changes of breast cancer organoids at 0, 24, 48 and 96 h after irradiation were observed to evaluate the irradiation-induced damage to the organoids. Results:Breast cancer cell lines and patient-derived tissues formed organoid structures at 6 d. HE staining showed the microstructures, and the expression profile of markers was spatially heterogeneous. The expression patterns of markers were similar between patient-derived organoids and original tumor tissues. Irradiation of MCF-7 breast cancer organoids led to growth arrested, and some of the formed organoids collapsed and the proliferating trend gradually recovered from 48 h to 96 h. MDA-MB-231 breast tumor organoids showed radioresistance, growth arrested, but the structures remained intact, the recovery trend was still not observed at 96 h. The tissue-derived organoids from triple-negative patients also showed radiation tolerance. After irradiation, the organoids continued to grow without significant structural changes, whereas the growth trend was significantly smaller than that in the non-irradiated group.Conclusions:Breast cancer organoids formed by in vitro culture of breast cancer cells from different sources and different molecular types have microstructure and heterogeneity, which can reflect the expression of source tissue markers and show different radioresistance. Organoids derived from triple-negative breast cancer are more resistant to irradiation.

ABSTRACT

OBJECTIVE:To study the m echanism of Liangfu pil ls in the tre atment of gastric ulcer. METHODS :Based on network pharmacology ,the active ingredients ,targets and gastric ulcer related targets of Liangfu pill were screened by searching TCMSP,GeneCards,OMIM and DisGeNET databases ,and the common targets of Liangfu pill and gastric ulcer were screened by Venny 2.1 software. The protein interaction network of Liangfu pill in the treatment of gastric ulcer was constructed by STRING database. Cytoscape 3.7.2 software was used to construct topological analysis of the network.The function of GO and the enrichment of KEGG pathway were analyzed by DAVID database. Active component-gastric ulcer target-KEGG pathway network was constructed for Liangfu pill. In animal experiment ,mice were randomly divided into blank group ,model group ,ranitidine group (positive control ,39 mg/kg),Liangfu pill low- ,medium- and high- dose groups (0.78,1.56,3.12 g/kg),and were given corresponding drugs intragastrically ,once a day ,for consecutive 7 days. After last administration ,except for blank group ,the other groups were given absolute ethanol (0.01 mL/g)intragastrically to induce the gastric ulcer model. After modeling ,the pathological changes of gastric tissue were observed and the gastric ulcer index was calculated ;the expression of TP 53,c-Jun,p38 MAPK,Akt,p-Akt and TNF-α protein in gastric tissues of mice were detected. RESULTS:A total of 9 active ingredients (i.g. isorhamnetin ,β-gsterol,kaempferol)and 166 common targets were screened in Liangfu pills. ABSTRACT

【Objective】 To investigate the clinical staffs′ knowledge about blood transfusion and their demands, so as to provide evidence for arranging the content for continuing education of blood transfusion medicine. 【Methods】 A self-designed questionnaire was used to investigate the blood transfusion related knowledge of 652 clinical staffs from other cities or counties/districts in Shaanxi Province, who came to our hospital to attend meetings, further training and study from July to August 2019. 【Results】 Clinical stsffs investigated lacked a deep understanding of blood transfusion and blood donation related knowledge, and even were ignorant of certain knowledge. The awareness rate of relevant knowledge was the highest among doctors, followed by laboratory technicians and the lowest among nurses. 【Conclusion】 The awareness rate of blood transfusion related knowledge among medical staffs is relatively low, suggesting the training of blood transfusion related knowledge should be enhanced.

ABSTRACT

OBJECTIVE:To explore the protective effect of Alpinia zerumbet extract on acute gastric ulcer model mice. METHODS:Totally 48 mice were collected and randomly divided into blank group ,model group ,positive group (Sanjiu weitai granules,20 mg/kg),A. zerumbet extract high-dose ,medium-dose and low-dose groups (2.34,1.17,0.59 g/kg,by crude drug ), with 8 mice in each group. They were given normal saline or relevant medicine intragastrically ,once a day ,for consecutive 7 d. Then,except for blank group ,other groups were given disposable intragastric administration of absolute ethanol 0.1 mL/10 g to establish acute gastric ulcer model. Another 48 mice were collected and grouped with same method ,and then given relevant medicine for consecutive 15 d. From the 10th day of administration ,except for blank group ,other groups were given intragastric administration of aspirin (20 mg/kg) for consecutive 6 d to establish acute gastric ulcer model. In ethanol induction model experiment,the formation of gastric ulcer and the index of gastric ulcer were observed in each group ;HE staining was used to observe the pathological changes of gastric tissue ;the levels of gastrin (GAS)in serum ,and the levels of SOD ,MDA and NO in gastric tissue were determined by ELISA. In the aspirin induced model experiment ,the formation of gastric ulcer and the index of gastric ulcer were observed and measured by the same method ;the levels of GAS ,TNF-α,IL-1β in serum,and the levels of PGE2 and COX- 2 in gastric tissue were determined by ELISA. RESULTS :Compared with model group ,obvious ulcer lesions were found in gastric tissue of rats in model group ;the levels of GAS ,TNF-α,IL-1β in serum,and MDA ,NO in gastric tissue were increased significantly , while the levels of SOD , PGE2, (No.QZYY-2019- COX-2 in gastric tissue were decreased si gnificantly(P<0.05 063). Compared with model group ,the degree of gastric lesions in administration groups was alleviated to some extent;gastric ulcer index was decreased to some extent , mail:997845460@qq.com while the levels of the above indexes in serum and gastric tissue were improved in varying degrees , with statistical 制。E-mail:672863283@qq.com significance in most indexes (P<0.05 or P<0.01). CONCLUSIONS:A. zerumbet extract exerts its protective effect on absolute ethanol and a spirin-induced acute gastric ulcer model mice,the mechanism of which may be relieving gastric mucosal injury through inhibiting oxidative stress and inflammatory response.

ABSTRACT

Objective:To observe the effects of different indomethacin anal suppository administration time on increased serum amylase, pancreatitis onset and the degree of pain after ERCP, and explore the optimal administration time.Methods:A total of 240 patients with cholangiopathy who were admitted and planned to be treated by ERCP in Department of General Surgery of Xinhua Hospital Affiliated to Shanghai Jiaotong University School of Medicine from May 2018 to May 2019 were randomly divided into 6 groups using random number method with 40 patients in each group, who were given 100 mg intrarectal indomethacin at 120 min, 60 min, 30 min before ERCP and 30 min, 60 min, 120 min after ERCP. The incidence of post-ERCP hyperamylasemia, post-ERCP pancreatitis (PEP) and the degree of postoperative pain were compared among different groups.Results:The increased times of blood amylase level and the incidence of hyperamylasemia and PEP at 24 hour, and postoperative 3 h and 24 h pain scores in preoperative medication group were significantly lower than those in postoperative medication group [(4.2±1.9) vs(4.7±2.1), 36.7% vs 46.7%, 19.2% vs 24.2%, (5.9±2.1) vs(6.4±1.4) and(3.8±1.7) vs (4.3±1.4)score, all P<0.05), and the amylase increased times (2.5±1.2), the incidence of hyperamylasemia and PEP[(27.5%(11/40) vs 12.5%(5/40), and postoperative pain score (3.0±1.2) were the least in preoperative 30 min medication group. Conclusions:Indomethacin anal suppository can reduce the incidence of postoperative hyperamylasemia and PEP and mitigate the pain degree after ERCP, and indomethacin given 30 minutes before ERCP may obtain the best preventive effect.

ABSTRACT

By removing a part of the structure, the tooth preparation provides restorative space, bonding surface, and finish line for various restorations on abutment. Preparation technique plays critical role in achieving the optimal result of tooth preparation. With successful application of microscope in endodontics for >30 years, there is a full expectation of microscopic dentistry. However, as relatively little progress has been made in the application of microscopic dentistry in prosthodontics, the following assumptions have been proposed: Is it suitable to choose the tooth preparation technique under the naked eye in the microscopic vision? Is there a more accurate preparation technology intended for the microscope? To obtain long-term stable therapeutic effects, is it much easier to achieve maximum tooth preservation and retinal protection and maintain periodontal tissue and oral function health under microscopic vision? Whether the microscopic prosthodontics is a gimmick or a breakthrough in obtaining an ideal tooth preparation should be resolved in microscopic tooth preparation. This article attempts to illustrate the concept, core elements, and indications of microscopic minimally invasive tooth preparation, physiological basis of dental pulp, periodontium and functions involved in tool preparation, position ergonomics and visual basis for dentists, comparison of tooth preparation by naked eyes and a microscope, and comparison of different designs of microscopic minimally invasive tooth preparation techniques. Furthermore, a clinical protocol for microscopic minimally invasive tooth preparation based on target restorative space guide plate has been put forward and new insights on the quantity and shape of microscopic minimally invasive tooth preparation has been provided.

ABSTRACT

Objective To investigate the effect of apatinib on the migration ability of nasopharyngeal carcinoma NPC cells after X-ray irradiation and involved protein expressions. Methods The migration abilities of human immortalized nasopharyngeal epithelial cells ( NP69) and nasopharyngeal carcinoma cells ( CNE-1, CNE-2 ) treated with different concentrations of apatinib ( 0, 5, 10 and 15 μmol/L) were compared by wound healing assay. The effect of apatinib on the activity of NPC cells was detected by CCK-8 for determining the suitable intervention concentration of apatinib. Then NPC cells were divided into control group, apatinib group (15 μmol/L), X-ray irradiation group and apatinib combined with X-ray irradiation group, and the migration ability of each group was compared by wound healing assay. The expressions of pVEGFR2, pSTAT3, STAT3, MMP-9 and EMT related proteins were detected by western blot. Results Compared with the NP69, the migration abilities of CNE-1 and CNE-2 were significantly enhanced ( t=-5. 759, -16. 578, P<0. 05) . Compared with the control group ( 0 μmol/L) , the migration ability of NPC cells after treatment with apatinib(5, 10 and 15 μmol/L) was significantly decreased in a concentration dependent manner ( t=2. 804-13. 362, P<0. 05) . Compared with the X-ray irradiation group, the wound healing rate of NPC cells in the apatinib combined with X-ray irradiation group was decreased ( t=5. 932, 2. 791, P<0. 05) , indicating that apatinib can significantly inhibit the migration of NPC cells after X-ray irradiation. Western blot assay showed that the expressions of pVEGFR2 and pSTAT3 were significantly decreased in NPC cells treated with apatinib, meanwhile, the expression of MMP-9 protein was significantly decreased, and the EMT-related protein was changed. Conclusions Apatinib inhibits migration of X-ray irradiated NPC cells by inhibiting EMT through down-regulating VEGFR2/STAT3/MMP-9 signaling pathway.

ABSTRACT

Three-dimensional cell culture is a novel type of in vitro culture method.This system can mimic the microenvironment of cell growth in vivo,where cells exhibit similar state and function to that in the in vivo environment.Currently,three-dimensional culture system has been widely applied in tissue engineering and tumor cell biology fields,etc.Radiotherapy is an important treatment of cancer.Radioresistance of tumor cells is the main cause of treatment failure,tumor recurrence and metastasis.Tumor cells can exhibit the resistant characteristics in the three-dimensional culture system,similar to those of tumor cells in vivo.Therefore,three-dimensional culture system can be adopted to investigate the radioresistance and underlying mechanism of tumor cells and identify the key factors regulating the radioresistance of tumor cells,which plays a pivotal role in enhancing the radioresistance and improving the effect of radiotherapy.This article will review recent research progress in the radioresistance and sensitization of tumor cells under three-dimensional culture system,aiming to provide reference for relevant investigations.

ABSTRACT

Objective To explore the relationship between MTHFR gene polymorphism and lung cancer in Han population of Heilongjiang Province.Methods Two hundred and twenty-five lung cancer patients were selected as the experimental group and the healthy subjects in the outpatient physical examination as the control group,A case-control study was used to analyze the association of MTHFR gene 677C/T,1298A/C SNP and lung cancer,and the gene typing was detected by Sanger double deoxidization chain termination method.Results In the control group,the frequencies of wild-type CC,mutant heterozygote CT,and homozygous TT genotypes of the MTHFR gene C677T were 34.2%,55.1%,and 10.7%,respectively.The frequencies of the three genotypes in the experimental group were 26.7%,50.2%,23.1%,respectively.The difference in the distribution of C677T SNP genotype frequencies of the experimental group and the control group was statistically significant (P =0.002),in which the mutation homozygous TT carriers were 2.78 times more likely to develop lung cancer than wild-type CC (OR(95%CI):2.78 (1.54 ~ 5.02),P =0.001);AA,AC and CC genotype frequencies ofthe A1298C locus of the MTHFR gene were 34.2%,55.1%,and 10.7%,respectively,and the control group was 64.0%.32.0%,4.0%,there was no significant difference between the two groups (P =0.247).The frequencies of AA,AC and CC genotypes in the A1298C locus of the MTHFR gene were 34.2%,55.1%,and 10.7%,respectively,and 64.0.%,32.0%,and 4.0% in the control group,respectively.There was no significant difference between the two groups (P).=0.247).The haploid analysis showed that the distribution frequency of TA haplotype in the experimental group was significantly higher than that in the control group (43.1% vs.35.3%).There was a statistically significant difference between the two groups (OR(95%CI):1.39 (1.06-1.81),P=0.016);while the frequencies of CC haplotypes in the experimental group and the control group were 10.6% and 17.1%,respectively.The difference was statistically significant (OR(95%CI):0.58 (0.39-0.85).P=0.005).There was a linkage disequilibrium between the two points of MTHFR gene 677 and 1298 (D'=0.48,P=0.003).The gene-environment interaction analysis of the MTHFR gene C677T polymorphism showed that based on the comparison between TT and CC genotype,age over 60 (OR(95%CI):4.0(1.78-9.32),P =0.001),male (OR (95%CI):5.55 (2.10-14.67),P=0.000),smoking (OR(95%CI):8.13 (2.29-28.85),P=0.000) and small cell lung cancer (OR (95%CI):1.28 (1.10-1.44),P=0.000) can increase the risk of lung cancer;based on the comparison between CT and CC genotype,women (OR(95%CI):2.09 (1.05-4.16),P=0.030),non-smoking population (OR(95%CI):2.43 (1.25-4.74),P=0.008) and small cell lung cancer (OR (95% CI):0.31 (1.16-1.59),P =0.000) can increase the risk of lung cancer.Conclusion MTHFR gene C677T is a genetic susceptibility gene for lung cancer and is associated with the risk of lung cancer.

Objective To study the value of the informatization clinical blood transfusion apps application in blood transfusion management under the new normal.Methods By retrospective analysis of the audit qualification rate of 16 988 general clinical blood transfusion and informatization clinical blood transfusion apps forms,and both types of analogy each stage time nodes correlation analysis.Results Review the qualification rate of the 8 616 general clinical blood transfusion form is 76.9%,the time of the doctor's advice to send is (20± 8) minutes,the time of the Blood Transfusion Department to receive is (90±48) minutes;review the qualification rate of the 8 372 informatization clinical blood transfusion apps form is 99.2%,the time of the doctor's advice to send is (3± 1) minutes,the time of the Blood Transfusion Department to receive is (2± 1) minutes.Conclusion Informatization clinical blood transfusion application form to adapt to clinical doctor operation,ensuring the safety of blood transfusion by shortening the each stage time nodes for clinical service.

ABSTRACT

Tooth bleaching has become a routine way of treating tooth staining. It is necessary to teach tooth bleaching in undergraduate education of Stomatology. The possibility of teaching bleaching was explored in our school and the teaching effects were assessed by questionnaires to undergraduate students of Stomatology in 2005 and 2006 degree after class. The outcomes indicated that a teaching mode of “3+3+2” made a positive effect on the teaching of tooth bleaching, combining with multimedia teaching,problem-based learning and bilingual teaching. The present article summed up the experience of teaching tooth whitening in undergraduate education of Stomatology and gave some advice for improving the quality of teaching.
